What is the renewal process for a ACME certificate in the Domino 12.0.1 CertMgr?
I see there is an "automatically renew" field in the TLS Credentials form.
Is CertMgr supposed to submit a new request to the ACME source/LetsEncrypt?

We have Automatically Renew set to 30 days and yes it renews your SSL cert automatically. We have it set up for our main Domino server, DEV server and Traveler server all from one Certificate Store database. Works great for us.

Certificate renewal process of Let's Encrypt using the certstore.nsf on the V12.0.x Domino server is automatic as per the "Certificate renew date" field value. The cert manager task (certmgr) on the Domino server will make sure it renews the certificates.
Example shows the renewal date and expiration date of the certificate.
On 1st Nov, 2022 the cert manager task on the Domino server will renew the certificates for the above host. Even before the 1st Nov, 2022. If you wanted to renew the certificate Today. We can just edit the document and change the Renewal date to Today's date. You can just type @Today it will have the Today date populated.
When you submit the request it will be processed and certificate will be renewed.
You can also change the global settings.
