The OEM version of IS Tuner is generic; it will work with any MSI package, as long as you have the vendor-supplied configuration file (*.itw) appropriate to the product ID you want to customize.
Each release of Notes 6.x includes a version-specific Tuner configuration file called LotusNotes.itw, in the same directory as the Notes MSI package. You can point Tuner to the correct .itw file every time you start it, but that’s a nuisance. You can avoid the annoyance by associating *.itw with “c:\Program Files\Lotus\InstallShield Tuner for Lotus Notes\System\iside.exe”, which lets you run Tuner with the appropriate configuration file by running the configuration file itself (LotusNotes.itw), either from a shortcut or from Windows Explorer.

I had the same problem and error message. It related to the Notes help for creating the transform file. The Notes help says get the file from the Apps/InstallShield Tuner on the CD, but that does not work. I found I had to get the file (In my case Lotus Notes 6.5.1) from the \Clients\W32Intel directory.
