Good Afternoon,
Each time I either compose a new email or reply to an email I have to always change my Delivery Options to set the reply to address, respond by date, expire date, Read Receipt request, Importance, Prevent Copying, etc. I even check the box that says, "Save these security options as the default."
Is there a way that I am able to actually check that box and it do something, like actually save those settings as the default?
Please let me know anyone and I will greatly appreciate it.

The Delivery options are being set when a mail is in edit mode (compose or reply being performed).
This will be specific to every mail depending upon what level of changes you are performing in "Delivery options" dialog box.
As far as your query is concerned, if you have checked the setting "Save these security options as the default.", then expected results is that while replying to any mail from now on, the "Delivery options" will show options "Sign" and "Encrypt" as checked by default.
Confirm if you are not getting this behavior too. I just tested and found that in every new mail options "Sign" and "Encrypt" as checked by default.

Click this box and the Sign and Encrypt options as the default options you have chosen will be saved as the default for all of your mail messages.

Other than "Sign" and "Encrypt" options which will be checked by default, you need to set other options everytime when you are working on mail like choosing the "Importance", "Delivery report", "Delivery priority" etc. as these option might vary for every mail that you receive.

This is correct. The "Save these security options as the default." will only save the settings under Security Options only. It will not affect other options like Importance, return receipt, etc.

Return Receipt and Spell check it can always be enabled for each messages you send via mail file preferences as shown below and these can be controlled via mail settings policy.
"Save these security options as the default" will only have Encrypt & Sign settings as these are considered as mail security options when sending emails. If you turn on Encrypt & Sign with option "Save these security options as the default" it is always enabled for each message you send next time. Additionally Encrypt and Sign email setting can also be controlled by Desktop settings policy.
These settings -> Importance, Delivery Priority, Prevent Copying and Mark Subject Confidential can't be enabled via policies. You may need to customize the mail template to have these set as default as per your requirement.
Coming to the Delivery Option - Advanced tab, these are specific to messages you send it. We can't set the Expiration Date and Reply by this date same as for all messages which you send in future.
Also, Reply to this memo should be addressed to is used when sender is going out and for his sent message the response should be notified to other person in the organization in his absence to have some action.
However, if your requirement is to have these enabled by default with some values you may need to customize the mail template as per your requirement. You may consult your in house Notes developer with these requirements.
Please note, if you customize the template and if you face any issues around the customization then, support will ask you to test it with standard template to understand if the issue is specific to customization. Also customization is not in the scope of support.
