We have downloaded en Evaluation version of lackberry Enterprise server 4.1. Do we need to have tie up with Mobile operator to test the application? can we test it without that?

In order to set up a successful BlackBerry environment, you need the BlackBerry devices themselves, some sort of connectivity for them, and a BlackBerry Enterprise Server.
Did you purchase some devices? If not, I’m unclear as to what you hope to test.
Generally, one of the cellular carriers will “loan” you a couple devices to test with (and, the devices would have connectivity) or they would allow you to purchase a couple devices with a 30-day return option.
Regardless, however, you need the device(s), connectivity and the server.
There ARE simulators, but those are really developer tools, not “does this work” tools …
