I have now upgraded to domino 8.5.2 and activated TL and DAOS.
I now backup all nsf databases with hp dp but this is taking to long time.
I have increased the size with around 100gb with daos but the backup still need to backup around 400gb data and this takes about 24h.
Do I need to backup all the files in the data folder for disaster recovery?

You need to use a special Domino Agent to backup Notes databases, provided by your backup software vendor, and if you have DAOS enabled, you need to have the version that supports DAOS.
Using a standard file backup is risky, especially with DAOS.
If you have transaction logging enabled, you also have to backup the transaction logs if you plan to ever do a restore from a full back + incrementals.

I backup the whole DAOS folder. Are you meaning in worst case scenario like disaster recovery?
Why do I need to backup the whole data folder?

You need to backup the entire data folder (eg C:\Program Files\IBM\Lotus\Domino\Data).
