I am failing to get an Domino 6.0.1 server to perform an LDAP query to Active Directory. I have this working OK at a different client. The only difference I can see is that this server has “Load Internet Configurations from Server\Internet Sites document” set to Enabled. LDAP is setup in Directory Assistance but when I enable it emails are held in mail.box until I disable Directory Assistance.
Does the server performing the LDAP lookup need to have a LDAP Site document created or is this only for another server quering Domino?

We had a similar problem with mail being held up in the mail.box. Come to find out that if the LDAP server that Domino is trying to connect to is down then mail will stop routing. Must be a Domino bug.
You are, however, on the right track. In order to have Domino connect to an Active Directory server you must have an LDAP document in Directory Assistance. Check to make sure that all the fields are correct and that the server name or ip is correct. Also make sure that it is trusted for credentials under the second tab.
