How can I do it? Even I empty the internet address field in the directory user profile, mail message will still sent to his/her mailbox. And I would like to have a message sent to the sender.

Subject: Auto-reply mail message for user already left
Are you talking about terminated user, or “left for the day” user?
If terminated, you delete the user from the Domino Directory, then the sender would get a " no such address" automatically. That is the standard way to handle this.
If you have some reason you need to leave the user profile in place, then perhaps you need to enable Out-of-Office (but that would deliver the message and respond to sender only once, and is best for long-term.)
OR create a Before-new-mail-arrives Agent that replies to the sender, including a copy of the original message, and then deletes the original message (so it never “appears” in the mailfile.)
